A comprehensive study of monkeys given the breast cancer drug letrozole reveals side effects that impact the brain. Published in JNeurosci, the research establishes the common marmoset as an important nonhuman primate model for studying the effects of estrogen-reducing treatments on the nervous system.
Letrozole is used to prevent breast cancer recurrence by interfering with the production of estrogens. Although side effects such as mood disturbances and memory issues have been reported in both humans and animals, little is known about how the drug impacts the brain.
Nicole Gervais and colleagues administered Letrozole to male and female marmosets via pudding for four weeks and observed many of the same behavioral changes, including hot flashes and increased anxiety, experienced by women receiving similar treatment. Letrozole also compromised the function of neurons in the hippocampus and impaired spatial memory. Together these findings emphasize the need for further investigation of breast cancer treatments and their effects on the brain.

كشفت دراسة حديثة، أجراها باحثون من جامعة تورنتو الكندية، أن عقارا يستخدم لعلاج سرطان الثدي، يمكن أن يؤدي إلى ضعف وظائف المخ لدى المرضى.
وقد نشرت نتائج الدراسة في العدد الأخير من دورية (Journal of Neuroscience) العلمية.
وأجرى الفريق دراسته لاكتشاف الآثار الجانبية لعقار “ليتروزول” (Letrozole)، الذي يستخدم على نطاق واسع لعلاج سرطان الثدي، على المخ.
وقد أجريت الدراسة على مجموعة من القرود المصابة بسرطان الثدي، رصد الفريق تأثيرات عقار “ليتروزول” على الجهاز العصبي لديها، ولاحظ فريق البحث أن عقار “ليتروزول” أثر على وظائف الخلايا العصبية في قرن آمون في الدماغ، وأدى إلى اختلال الذاكرة المكانية.
وقالت الدكتورة نيكول جيرفيه، قائد فريق البحث، إن “نتائج الدراسة، تدفعنا إلى إجراء مزيد من الدراسات الأخرى للتحقق من علاجات سرطان الثدي التي تستهدف هرمون (الإستروجين) لرصد آثارها على الدماغ”.
ويستخدم عقار “ليتروزول” لمنع عودة انتشار سرطان الثدي عن طريق التدخل في إنتاج هرمون “الإستروجين” الذي يؤدي عرقلة إنتاجه لمنع انتشار أورام الثدي.
ووفقا للوكالة الدولية لأبحاث السرطان، التابعة لمنظمة الصحة العالمية، فإن سرطان الثدي هو أكثر أنواع الأورام شيوعا بين النساء في جميع أنحاء العالم، ومنطقة الشرق الأوسط خاصة، إذ يتم تشخيص نحو 1.4 مليون حالة إصابة جديدة كل عام، ويودي بحياة أكثر من 450 ألف سيدة سنويا حول العالم.
